Multi-color Multi-photon Imaging

Average projection image of a stack of optical slices taken through 25um of a Convallaria sp. rhizome. Fluorescence detection channels were centered about 500nm (green) and 650nm (red) wavelengths. The composite image indicates the differential emission of auto-fluorescence from different cellular constituents stimulated with multi-photon absorption of laser pulses centered about 780 nano meters wavelength. Multi-photon imaging of systems with multiple fluorochromes in a spectral detection system often proves advantageous because of reduced interference from reflected cw light, pinholes, and non-optimal dichroic mirrors.
